I completely forgot that Feifei had/has a Xanga. I read some of her really old posts, like the ones about Faustus and college apps. Ah, nostalgia.
I have this strong urge to finish the Katobe drawing I started but I can't very well do that here at work. Grrr. If I had a freakin' pencil, I'd doodle. I don't like drawing in pen 'cause I tend to be a bit of a perfectionist when it comes to drawing.
Episode 27 of Kamen Rider Kabuto will have lots of Kazuki! Yay! And he gets another change of clothes. Exciting.
Western blots take too long.
1) Protein assay: >30 mins
2) Loading: ~10-20 mins
3) Running: ~1 hr
4) Transfer preparation: ~10-30 mins
5) Transferring: >1 hr
6) 1st wash: 3x 5 mins
7) Blocking: 1 hr
7b) Optional wash: 3x 5 mins
8) Primary antibody: overnight in cold room or 1 hr at room temp
9) 2nd wash: 3x 5 mins
10) Secondary antibody: 1 hr
11) 3rd wash: 3x 5 mins
12) ECL solution: ~1-5 mins
13) Developing: ~1-60 mins (depends on the protein you're trying to detect)
After all of that, you finally get to see whether you've fucked up your experiment or not. >.< It took about 6 gels for my boss to figure out that something may be wrong with our lysis buffer. It's too much work.
